Dr Kassen's 8-month Blood Sugar and Lipid Profile Snapshot

This episode continues discussing my personal health journey and data from August 2024 to May 2025 (including limited data from March 2024), focusing on my prediabetes and associated lipid profile changes as updated on 7 May 2025.

The analysis reveals a successful reversal of prediabetes within three months (September to December 2024) through significant lifestyle and dietary modifications, primarily intermittent fasting (IF) and a low-carbohydrate diet (Low-carb). While blood glucose control remained stable within the normal range even with some dietary liberalisation and a cessation of intermittent fasting (IF), the lipid profile showed some increases in total and LDL cholesterol between September 2024 and December 2024, but did drop back down into the normal ranges for May 2025. Thus, to date, key lipid ratios are indicative of a very favourable cardiovascular risk (QRisk2 score).

Taken together, this highlights the close interplay between insulin resistance (IR), blood sugar control, and lipid metabolism, and underscore the importance of continued monitoring and a sustainable healthy lifestyle.

Healthspan, Lifespan, Disease Trends, and Economics: A Global Overview

While global average lifespans are increasing, major disparities exist both in terms of lifespan and healthspan, and these disparities are likely to continue in the future, with major regional variations in life expectancy, healthcare access, disease burden, and its socio-economic impact. The modelling for the future requires caution given the modelling methods used, but nevertheless provides an overview of likely future scenarios. These findings point towards a need for both global efforts to reduce the health gap, and regional-specific interventions tailored to different challenges. A few sources for those interested in delving a bit deeper: Global Healthspan-Lifespan Gaps Among 183 World Health Organization Member States; https://www.healthdata.org/news-events/newsroom/news-releases/global-life-expectancy-increase-nearly-5-years-2050-despite; COVID-19 eliminated a decade of progress; World health statistics 2024; Burden of disease scenarios for 204 countries and territories, 2022–2050: a forecasting analysis for the Global Burden of Disease Study 2021; https://www.visualcapitalist.com/japans-aging-problem-in-one-chart/; Longevity leap: mind the healthspan gap

My Prediabetes Journey: IR and T2DM - A Global Health Crisis_Part 4

Insulin resistance, type 2 diabetes, and metabolic syndrome are major and escalating global health problems. Key factors driving these issues include obesity, unhealthy lifestyles, and genetic predispositions. These conditions place a significant burden on individuals and healthcare systems due to morbidity, mortality, and healthcare costs. Effective prevention and management strategies rely on lifestyle modifications, early diagnosis and, in some cases, medication. The continued collection of data and further research on the underlying mechanisms will be essential to combat these complex challenges effectively.

My Prediabetes Journey: T1DRA Study_Part 3

Today we discuss an experience I had with an at-home blood sample collection process for the T1DRA study, a UK-based research initiative investigating type 1 diabetes risk factors in adults. The study aims to identify individuals at increased risk and improve understanding of the condition's development. Participants aged 18-70 provide blood samples, and those with multiple risk markers receive information on type 1 diabetes management and clinical trials. The accompanying video is available on YouTube here and details the sample collection procedure, offering timestamps for viewers' convenience. The study seeks diverse participation to ensure representative findings for the general UK adult population. An unboxing of the study material received can be accessed here.

Although my concerns are about acquiring T2DM, I felt it necessary to partake in this T1DM study. Links available on my YouTube channel.

Cystic Fibrosis (CF): Metabolic Adaptations of Pseudomonas Aeruginosa_Part 2

Cystic fibrosis (CF) is a complex genetic disorder with significant implications for those affected. While there is currently no cure, advancements in our understanding of the disease, its genetic underpinnings, and the development of targeted therapies are improving both life expectancy and quality of life for those affected. Yet, many challenges remain especially on the metabolic adaptations of Pseudomonas aeruginosa (P. aeruginosa) during chronic infections in the lungs of individuals with Cystic Fibrosis (CF). These adaptations are not simply a consequence of the environment, but an active process driven by specific mutations and metabolic shifts that impact virulence, immune evasion and interactions within the CF lung. A key finding is that mutations in the PDHc, causing shifts in pyruvate metabolism, play a central role in driving this adaption. Further research into these metabolic changes is essential for developing more effective treatment strategies for CF-related infections. The use of advanced metabolomics, proteomics, and in vitro models is essential to understanding this dynamic process.

X-ray Crystallographic MRes & PhD Research: Pre-DeepMind AlphaFold_Part 1

The convergence of AI and structural biology is revolutionising how researchers investigate complex biological and biomedical problems. By applying these techniques to their research, scientists are closer to developing novel therapeutic strategies and preventative measures. While AI offers an immense acceleration to the generation of hypotheses and models, it should not be seen as a replacement for traditional experimental techniques and the valuable insight that they bring.

NB. My research (MRes and PhD) at Imperial College London during 2012 to 2021 centred around protein crystallisation methodologies and x-ray crystallography, which aided 3-D protein structures determination. Data and information gained from crystallography is vital to the success of rational drug design and medicinal therapeutics and biotechnological applications. If AlphaFold existed in its current form during parts of my research period, I'm positive it would have helped accelerate the painstaking research we conducted and potentially helped boost our findings and results. Sleep Patterns and Chronic Disease Risk

Dr Sean's YouTube video discusses a study published in Nature Medicine examining the link between sleep patterns and chronic diseases. The study, using data from wearable devices like Fitbits, found significant associations between poor sleep quality, quantity, and regularity, and conditions such as obesity, hypertension, and depression.

Researchers employed long-term data from a large US research program, offering a more comprehensive understanding than previous studies. The video also recommends additional resources on sleep from the National Institute of Neurological Disorders and Stroke (NINDS).

My Prediabetes Journey: Diagnosis_Part 1

Part 1 - This episode is a podcast style interaction by two AI generated hosts talking about blood sugar control as well as other markers tested in my blood work. They did a great job bringing it to life, don't you agree. Here, they summarise and discuss key findings from my full blood test and prediabetes diagnosis confirmed on 5 September 2024 after a proactive approach to my NHS GP Surgery. For a more detailed understanding, head over to my YouTube Channel @DrSean-888 where I talk about my blood test and results and the reasons behind it all. Follow me and subscribe if interested. More content to come, including follow-up episodes on my health journey and goals. I also post on Instagram: @sckvvv_drathome. Thank you for listening 🎶.
