EP7|Microfluidic mRNA製程革新:實現疫苗量產與品質雙贏

Significance 研究意義

本研究提出以矽-玻璃製成的 SCALAR ( Silicon Calable ALipid RNP) 微流體晶片,將單一混合單元無縫擴增至 1×、10×、256×,使 mRNA-LNP 產量從 0.1 L /hr 躍升至 17 L /hr,同時保持粒徑 (~70 nm)、封裝效率 (>85 %) 及體內免疫效力一致。

在 COVID-19 疫情中,LNP 配製是疫苗產線瓶頸;該平台證明可在不改變配方參數的情況下,直接由實驗室規模過渡到臨床與商業量產,顯著縮短反應新興病原的時程。

The study introduces a silicon–glass SCALARmicrofluidic platform that parallelizes identical mixing units (1×, 10×, 256×),boosting mRNA-LNP output from 0.1 L /hr to 17 L /hr while preserving particlesize (~70 nm), encapsulation efficiency (>85 %), and in-vivo potency.

During the COVID-19 pandemic, LNPformulation was a production bottleneck; SCALAR enables seamless scale-upwithout re-optimizing formulation parameters, accelerating the response toemerging pathogens.

 

Importance 關鍵重要性

製程韌性:晶片可耐 >1,000 PSI、高溫滅菌並可重複清洗 (非離子界面活性劑或高壓蒸氣),克服 PDMS 與 COC 一次性耗材易溶脹或汙染的限制。

產能與成本:256× 晶片每小時可生產約 34,000 劑 (0.5 mL/劑) 疫苗;若採 12-inch 晶圓,可進一步堆疊 >1,000 個混合單元,單片成本估計 < US$100。

品質一致性:自動化壓差驅動與階梯-樹枝流道設計確保各通道流阻均一,實驗顯示 90 % 以上混合長度於三種規格晶片皆無顯著差異。

Process robustness: Silicon–glass chipstolerate >1,000 PSI, autoclaving, and surfactant cleaning, overcomingswelling/leaching issues seen with PDMS or COC disposables.

Throughput & economics: A 256× chipyields ~34 k vaccine doses /hr; scaling to 12-inch wafers (>1,000 units)could drop per-chip cost below US $100.

Product consistency: A ladder-plus-brancharchitecture equalizes flow resistance; fluorescence-dye studies show identical90 % mixing lengths across 1×, 10×, and 256× chips.

Continuous manufacturing & cold-chainrelief: SCALAR can be integrated with inline dialysis or TFF, enablingend-to-end continuous flow and point-of-demand manufacturing that mitigatescold-chain constraints in low-resource settings.-

📌Significance :The significance of this study lies in exploration of potential therapeutic agents for non-alcoholic fatty liver disease (NAFLD) and metabolic syndrome, that are increasingly prevalent due to rising obesity rates. The study investigates the effects of cannabinoids, specifically THCV and CBD, for their potential to improve insulin sensitivity and lipid metabolism. This could lead to new pharmacological treatments for NAFLD, as effective therapies.

📌Importance of lipidology :The effects of THCV and CBD on lipid levels were examined in a variety of in vitro and in vivo systems, with special emphasis on models of hepatosteatosis. Treatment also induced post-translational changes in a variety of proteins such as CREB, PRAS40, AMPKa2 and several STATs indicating increased lipid metabolism and, possibly, mitochondrial activity. They show here that both THCV and CBD reduce intracellular lipid levels in vitro in a model of hepatosteatosis and adipocytes, mobilize zebrafish embryo lipid yolk stores and inhibit hepatosteatosis in obese mice.
